Targeting Systems offers expression vectors and assay reagents for Cypridina Luciferase, an ultrasensitive secreted luciferase from a marine Ostracod. The new luciferase uses Cypridina luciferin as substrate, which is distinct from firefly luciferin (substrate for firefly luciferase), and coelenterazine (substrate for Gaussia and Renilla luciferase). Targeting Systems already offers a significantly improved, brighter green-emitting Renilla luciferase mutant and Gaussia luciferase,(the brightest secreted luciferase). The addition of Cypridina luciferase enables analysis of multiple promoter activities in the same group of transfected cells. The Cypridina assay reagent is amenable to single cell assays on live cells for use in downstream applications. The enzyme is fairly stable and samples can be stored frozen for months without loss of activity. Cypridina luciferase offers flexibility, as a secreted or intracellular reporter, and significantly reduced assay time, because there is no need for cell lysis.
