Generating induced pluripotent stem (iPS) cells can be challenging because viral delivery of reprogramming factors often results in low reprogramming efficiency. By using doxycycline-inducible lentiviral expression, you can increase reprogramming efficiency over traditional, non-induced viral expression systems (Brambrink et al., 2008). Each doxycyline-inducible lentivirus in this set expresses one of four mouse transcription factors (Oct4, Sox2, Klf4, or c-Myc), GFP, and the reverse tetracycline transcriptional activator (rtTA) and has been proven to reprogram MEF’s to iPS cells.
