Description

The dominant function of mitochondria is the production of more than 90% of the cell's energy in the form of ATP through oxidative phosphorylation (OXPHOS). The OXPHOS system is comprised of five multimeric enzyme complexes (complex I-V). Two mobile carriers, ubiquinone and cytochrome c, are used to transfer electrons from NADH, produced by the oxidation of carbohydrates and lipids, to molecular oxygen. This creates a proton electrochemical gradient that is harnessed by Complex V to synthesize ATP.

Mitochondrial dysfunction is associated with multiple diseases, such as cancer, diabetes and neurodegenerative diseases. Furthermore, toxic effects of certain drugs are due to those drugs causing mitochondrial dysfunction. In a study using EMD Millipore's MILLIPLEX MAP OXPHOS Panels for multiplexed quantitation of OXPHOS complexes, several drugs were examined for their effects on mitochondrial complex levels.

MILLIPLEX MAP Human OXPHOS Magnetic Bead Panel (Cat. No. H0XPSMAG-16K) is available as a premixed panel and is to be used for the simultaneous detection of Complex I, II, III, IV, V, and NNT in human cell lysate or tissue extract.
MILLIPLEX MAP Rat/Mouse OXPHOS Magnetic Bead Panel (Cat. No. RM0XPSMAG-17K) is available as a premixed panel and is to be used for the simultaneous detection of Complex I, III, and V in rat/mouse cell lysate or tissue extract.

The reagents and sample preparation procedures for the MILLIPLEX MAP OXPHOS Magnetic Bead Panels (both Human and Rat/Mouse) have been optimized to keep the OXPHOS complexes intact, thus maintaining their enzymatic activities. The antibody pairs were carefully selected to bind to the different subunits of each complex, ensuring specific detection of the intact OXPHOS complexes rather than any dissociated subunits. The resulting panels allow you the convenience of quantitative detection of multiple intact OXPHOS complexes from cell lysates or tissue extracts in a single well or a 96-well plate.
