Heparin-binding EGF-like Growth Factor (HB-EGF) is an EGF family growth factor that is synthesized as a transmembrane precursor (proHB-EGF). ProHB-EGF, which contains one EGF-like domain, is subject to variable O-linked glycosylation and N-terminal truncation. In human, this precursor is the cellular receptor for diphtheria toxin. ADAM12 or ADAM17-mediated cleavage of proHB-EGF releases a 22 kD soluble HB-EGF; the cytoplasmic fragment functions as a transcriptional repressor.
