Hello, I am new to culturing cells of this nature. Do you know any helpful links to specific sites that not only provide the "standard" protocol for Culturing MCF 7 Cells like at the ATCC site, but "how much" of the culture medium is needed. My superior wants me to write up a "very" detailed protocol of the cell culturing medium before I can start work with the MCF-7 Cells. Anything would be helpful. Thanks!
The minimum amount of culture medium is simply that which will completely cover the bottom of the dish or flask. Of course, given that the cells will be metabolizing the nutrients in the medium and secreting wastes, you will have to change the medium more often if you only use the minimum amount. Using too much medium is wasteful. As a rule of thumb, 10 ml for a 10 cm diameter dish is a good amount. Depending on the % confluence and growth rate of the cells, changing the medium every 3 days or so should work well. BTW, these guidelines are true for essentially any adherent cell line, not just MCF-7 cells.
