Does anyone know what's the easier jurkat cell line to transfect? I'm trying to transfect JKHM but it haven't worked yet.
Although there's not too much in the literature about that line, there are many references to other Jurkat lines that have been transfected with various constructs. Before you start trying other Jurkat lines, are you sure that everything is working with the transfection? What exactly do you mean by "it doesn't work"? Do the cells die or do you get no protein made? Do you have antibiotics in the medium? Is the construct correct? Have you titrated the transfection reagent, DNA concentration, transfection time etc.? Have you tried electroporation or nucleofection (Amaxa)? The point is that there are many variables to consider when transfecting a cell line, any of which could render the transfection unsuccessful. For example, if the transfection reagent concentration is too low, you won't get much, if any, DNA into the cells. If it's too high, the cells will die. There are as many variables to consider with electroporation techniques as well.
