DNA ligases join linear DNA fragments together via covalent bonds. DNA ligation involves creating a phosphodiester bond between the 3' hydroxyl group of one nucleotide and the 5' phosphate of another. Ligation of DNA fragments is an essential step in many molecular biology techniques, including gene cloning and messenger RNA (mRNA) fingerprinting. For efficient ligation, DNA strands must be prevented from self-ligating (self-circularization and concatenation) by dephosphorylation of DNA ends. The enzyme used to ligate DNA fragments is T4 DNA ligase, originally isolated from the T4 bacteriophage. This enzyme will ligate DNA fragments that have blunt ends or overhanging, cohesive ends that are annealed together.
