    The Auto 2000 utilizes the AO/PI staining method (Acridine Orange / Propidium Iodide) for identification of live and dead nucleated cells. The AO dye stains the DNA in all nucleated cells in order to differentiate nucleated cells from debris and unwanted cell types, such as platelets red blood cells. The PI dye stains the DNA in nucleated cells with compromised cell membranes, allowing for identification of dead nucleated cells. The ability to accurately analyze primary cells within samples containing varying amounts of red blood cells eliminates the need to lyse samples, simplifying the analysis process and reducing variability due to processing. With the traditional trypan blue method, non-nucleated cells can be incorrectly counted, yielding viability calculations that are too high.

    Simple, Automated Analysis

    The Auto 2000 features a user-friendly touch screen with pre-set assays for PBMCs, nucleated cells, stem cells, and other cell types. Simply pipette 20µl of stained sample into a disposable counting chamber, insert into the Auto 2000, select the appropriate assay, and click count. In less than 30 seconds, the Auto 2000 acquires and analyzes bright field and fluorescent images and displays live / dead cell counts, concentration, mean diameter, and percent viability.

    The proprietary Cellometer software is optimized for accurate analysis of cells in clumps and irregular-shaped cells. User-specific assay protocols with customized counting parameters can be created for unique cell types. Automated analysis using tested mathematical algorithms eliminates the subjectivity and inter-operator variability associated with manual cell counting.
