    Fast(er) PCR

Can you complete a PCR run in less than 30 minutes? If not, check out “fast PCR,” a term coined to refer to a standard PCR protocol that underwent a make-over for speed—namely, faster temperature changes, faster enzymes, and more reducing or combining of steps in the protocol. (See Biocompare for an illustrated tutorial.)

    Genotyping

To paraphrase The Naked City, there are three billion base pairs in the human genome; each one tells a story. Yet for the geneticists whose job it is to map phenotypic traits to genomic addresses, sifting through...

Methylation Sensitive Hi-Res Melting Using Calibration

Bisulfite conversion of genomic DNA enables amplification-based detection of 5-methylcytosines. Methylation-sensitive (MS) Hi-Res Melting is a fast and simple detection method that compares melting temperature (Tm) and melting curve shape to assess methylation in a region of interest. Individual or multiple CpGs can be analyzed. Using the High Sensitivity Master Mix, with internal calibration, this technique is sensitive enough to show a single methylation event within a group of CpGs. High discrimination is possible because of the exquisite sensitivity of Hi-Res Melting combined with the internal calibration made possible with HSMM.

Small Amplicon Genotyping Using The LightScanner® System

Small amplicon genotyping is a simple yet powerful genotyping technique. No probes are required, making this technique rapid and inexpensive. Until recently, without probes, accurate closed-tube genotyping has been difficult. Now, with the high-resolution LightScanner instrument and a specialized genotyping master mix containing LCGreen® Plus dye and internal calibrators, small amplicon genotyping can produce the same high accuracy results associated with probe-based techniques.
